- - - - - - Important information. Please read carefully. FENFEDRIN TM Capsules Relieves nasal congestion in common colds and allergic rhinitis (eg. hayfever). COMPOSITION Each capsule contains Chlorpheniramine Maleate Pseudoephedrine Hydrochloride 4mg 60mg FE0201/FE0202-004 11.21 Pharmacodynamics Chlorpheniramine Maleate belongs to the alkylamine antihistamines, which are among the most active H1-blockers and are generally effective in relatively low doses. It is used to reduce rhinorrhoea and sneezing. Pseudoephedrine is a sympathomimetic agent with direct and indirect effects on adrenergic receptors. It is said to have less pressor activity and CNS effects than ephedrine of which it is a stereoisomer. Pseudoephedrine hydrochloride is used as a nasal and bronchial decongestant. Both active ingredients are readily absorbed from the gastro-intestinal tract. Pseudoephedrine is resistant to metabolism by monoamine oxidase and up to about 90% of a dose in 24 hours is largely excreted unchanged in the urine. Less than 1% is excreted as nor-pseudoephedrine which is pH dependent. The half-life of several hours is reduced in acid urine. Chlorpheniramine is extensively metabolised to polar and non-polar metabolites. The excretion of the unchanged drug is increased by acidification of the urine and increased flow of urine. After daily oral administration the forms excreted are about 20% unchanged drug, 20% as monodesmethylchlorpheniramine, 5% as didesmethylchlorpheniramine. Less than 1% is excreted through the faeces.
