Date of Revision WHAT FEPRIL SYRUP 120MG/5ML IS USED FOR Fepril Syrup 120mg/5ml is used for relief of fever and mild to moderate pain associated with conditions such as aches due to headaches, colds and flu symptoms, teething pain, toothaches, earache and post vaccination reactions. HOW FEPRIL SYRUP 120MG/5ML WORKS Fepril Syrup 120mg/5ml has pain relief and anti-fever properties. It works by blocking the pain impulse generation and increasing the pain threshold. It also increases the blood flow across the skin, heat loss and sweating to reduce pain. BEFORE YOU USE FEPRIL SYRUP 120MG/5ML _-When you must not use it _ You must not take this medicine and should talk to your doctor or pharmacist if: • You are allergic to Paracetamol or Acetaminophen or any other ingredients found in the syrup. • You are not sure whether you should start taking this medicine _Pregnancy and breast-feeding _ Do not use if you are pregnant, thinking of becoming pregnant, or breast feeding. FEPRIL SYRUP 120MG/5ML DESCRIPTION A pinkish red clear syrup with a raspberry flavor. COMPOSITION Each 5 ml contains Paracetamol :120mg _Preservatives:_ Methyl Paraben :6 mg Propyl Paraben :1 mg PHARMACODYNAMICS _Pharmacotherapeutic _ _group_: Other Analgesics and Antipyretics; anilides _ATC Code_: N02BE01 Paracetamol is a centrally acting analgesic and antipyretic with minimal anti-inflammatory properties. Analgesic The mechanism of analgesic action has not been fully determined. Paracetamol may act predominantly by inhibiting prostaglandin synthesis in the central nervous system (CNS) especially cyclooxygenase (COX)-2 and, to a lesser extent, through a peripheral action by blocking pain impulse generation. The peripheral action may be due to inhibition of prostaglandin synthesis or to inhibition of the synthesis or actions of other substances that sensitize pain receptors to mechanical or chemical stimulation. Antipyretic Paracetamol probably produces antipyresis by acting centrally on the hypothalamic heat-regulating center to produce peripheral vasodilation resulting in increased blood flow through the skin, sweating, and heat loss. The central action probably involves inhibition of prostaglandin synthesis in the hypothalamus. PHARMACOKINETICS Paracetamol is rapidly and almost completely absorbed from the gastrointestinal tract following oral administration. Approximately 90 to 95% of a dose is metabolized in the liver, primarily by conjugation with glucuronic acid, sulfuric acid and cysteine. An intermediate metabolite, which may accumulate in overdosage after the primary metabolic pathways become saturated, is hepatotoxic and possibly nephrotoxic. The half-life of paracetamol, 1 to 4 hours following oral administrations, does not change with renal failure but may be prolonged in acute overdosage, in some forms of hepatic disease, in the elderly, and in the neonate.
