PHARMACOLOGY:
Albendazole’s mode of actionhas been shown to be based on the
inhibition of the uptake and efficiency of utilization of
glucose by roundworms and tapeworms and similar activity is presumed
in liver fluke
Albendazole mode of action is to inhibit energy metabolism by
uncoupling phosphorylation reactions. In fasciola
hepatica, cellular changes due to Alebendazole include deformed
mitochondria, dilatation of the rough endoplasmic
reticulum, vesiculation of the golgi field and a reduction or absence
of secretary granules.
PHARMACOKINETICS:
The pharmacokinetics of albendazole and the same as when the products
are administered separately. Peak plasma
levels are attained 15 hours after treatment. The depletion curve is
linear and indicates a drug half-life 10 hours.
Albendazole absorbed from the gut and rapidly metabolized in the liver
almost completely to the 2- amino sulfone
metabolite plus 3 o 4 other metabolites. Peak plasma albendazole
levels are reached within 8 to 24 hours. Due to
plasma binding, levels persist which are adequate to exert a sustained
anthelminitic effect on Haemonchus contortus.
Tissue residues, however, are minimal.
Albendazole is metabolized primarily in the liver and, due to
concentration in the bile, excretion of metabolites is largely
via the faecal route.
INDICATIONS:
Control of susceptible mature and immature gastrointestinal
roundworms, large lung worms, liver fluke, nasal bots;
reduces output of viable worm and fluke eggs and provides sustained
control of barber’s pole worm in sheep.
Control of the following albendazole sensitive stomach. intestinal and
bowel worms: barber's pole worm (Haemonch us
contortus), stomach hair worm (Trichostrongylus axei), black scour
worm (Trichostrongylus sp.), small brown
stomach worm (Ostertagia sp.),
