Manufacturer and Product Registration Holder 10. Date of revision WHAT _VALTHOTAN_ IS USED FOR Valsartan is used to treat high blood pressure (hypertension), heart failure and post- heart attack. HOW _VALTHOTAN_ WORKS Valsartan is an angiotensin II receptor antagonist. Valsartan keeps blood vessels from narrowing, which lowers blood pressure and improves blood flow. BEFORE YOU USE _VALTHOTAN_ - _When you must not use it _ Do not use take _Valthotan_ if - You are allergic to valsartan or to any of the excipients in this product. - You are pregnant. _Pregnancy and lactation _ _ _ Do not take _Valthotan_ if you are pregnant, trying to get pregnant or think you may be pregnant. Do not take _Valthotan_ if you are breast- feeding. Ask your doctor or pharmacist for advice before taking any medicine. - _Before you start use it _ To make sure you can safely take valsartan, tell your doctor if you have any of these other conditions: If you have a serious kidney disease or are undergoing dialysis If you have a liver disease If you are suffering from heart failure or have experienced a heart attack If you are suffering from vomiting or diarrhea or are taking high doses of a diuretic (water pill) If you are already taking an ACE- inhibitor If you ever had a swelling mainly of the face and throat while taking other drugs (including an ACE-inhibitor). - _Taking other medicines _ Tell your doctor or pharmacist if you are taking or have recently taken any other medicines, including medicines obtained without prescription. Tablet 80mg, it occurs as a pale red film coated, round biconvex tablet; engraved with “1007” on one surface and “S || D” script and score on the other surface. For Valthotan F.C. Tablet 160mg, it occurs as a dark yellow film coated tablet, are oval-shaped with beveled edges, and engraved with “S || D” script and score on one surface and engraved with “736” script on the other surface. PHARMACODYNAMICS Angiotensin II antagonist. The active hormone of the renin-angiotensin-aldosterone system (RAAS) is angiotensin II, which is formed from angiotensin I through ACE. Angiotensin II binds to specific receptors located in the cell membranes of various tissues. It has a wide variety of physiological effects, including in particular both direct and indirect involvement in the regulation of blood pressure. As a potent vasoconstrictor, angiotensin II exerts a direct pressor response. In addition, it promotes sodium retention and stimulation of aldosterone secretion. Valsartan is an orally active, potent and specific angiotensin II (Ang II) receptor antagonist. It acts selectively on the AT 1 receptor subtype, which is responsible for the known actions of angiotensin II. The increased plasma levels of Ang II following AT 1 receptor blockade with valsartan may stimulate the unblocked AT 2 receptor, which appears to counterbalance the effect of the AT 1 receptor. Valsartan does not exhibit any partial agonist activity at the AT 1 receptor and has much (about 20,000 fold) greater affinity for the AT 1 receptor than for the AT 2 receptor.
