PROPERTIES
PHARMACODYNAMICS
Glucophage is an antidiabetic medicine that
belongs to the group of biguanides. Metformin,
the active ingredient in Glucophage, reduces
hepatic glucose production, increases insulin
sensitivity in muscles and delays intestinal
glucose absorption.
A reduction of diabetic complications has been
shown in overweight type 2 diabetic adult
patients treated with Glucophage as first-line
therapy after diet therapy. Glucophage is
associated with either a stable body weight or
modest weight loss.
PHARMACOKINETICS
Absorption:
After an oral dose of metformin hydrochloride
tablet, maximum plasma concentration (C

is reached in approximately 2.5 hours (t

between 1.5 and 3.5. Absolute bioavailability
of a 500 mg or 850 mg metformin
hydrochloride tablet is approximately 50-60%
in healthy subjects. After an oral dose, the non-
absorbed fraction recovered in faeces was 20-
30%.
After oral administration, metformin
absorption is saturable and incomplete. It is
assumed that the pharmacokinetics of
metformin absorption is non-linear.
