Date of revision WHAT YSP ALLOPURINOL TABLET IS USED FOR It is used to prevent gout attack and reduce the formation of excessive uric acid levels. However, it is not used to treat established attacks of gout. HOW YSP ALLOPURINOL TABLET WORKS Allopurinol is a xanthine-oxidase inhibitor agent which reduces the formation of excessive uric acid. BEFORE YOU USE YSP ALLOPURINOL TABLET - _When you must not use it _ Do not take this medicine if: • you are allergic to allopurinol or any other ingredients of this medicine (listed in Product Description) _ _ - _Before you start to use it _ _Pregnancy and lactation _ Please consult your doctor or pharmacist if you are pregnant, planning for pregnancy or breast-feeding before using any medicine. - _Taking other medicines _ Tell your doctor if you are taking any other medicines, including any that you buy without a prescription from a pharmacy, supermarket or health food shop. HOW TO USE YSP ALLOPURINOL TABLET - _How much to use _ The recommended dose for adult is to start with 100 mg once daily. The following dosage schedules are suggested: 100 to 200 mg daily in mild conditions, 300 to 600 mg daily in moderately severe conditions 700 to 900 mg daily in severe conditions. If the daily dosage exceeds 300 mg and develop gastrointestinal intolerance, a divided doses regimen may be appropriate. Follow all directions given to you by your doctor and pharmacist carefully. Allopurinol may possibly enhance effects of anticoagulants such as nicoumalone and warfarin. Plasma-cyclosporin concentration possibly increased by Allopurinol and results in increased risk of nephrotoxicity. The plasma half-life of adenine arabinoside is increased in the presence of allopurinol. When two products are used concomitantly,extra vigilance is necessary to recognise enhanced toxic effects. Oxipurinol, the major metabolites of allopurinol and itself therapeutically active, is excreted by the kidney in a similar way to urate. Hence, drugs with uricosuric activity, eg. probenecid or large doses of salicylates may accelerate the excretion of oxipurinol. This may decrease the therapeutic activity of allopurinol, but the significance needs to be assessed in each case. If allopurinol is given concomitantly with chlorpropamide when renal function is poor, there may be an increased risk of prolonged hypoglycaemic activity. Allopurinol may inhibit hepatic oxidation of phenytoin, but the clinical significance has not been demonstrated. It is a potent xanthine oxidase inhibitor with the chemical formula 4-hydroxypyrazole (3,4-d) pyrimidine (HPP).
