FRUSEMIDE-CLARIS Claris Lifesciences Pty Ltd NAME OF THE MEDICINE Frusemide-Claris, Solution for Injection, for Intramuscular (I.M.) or Intravenous (I.V.) use. Frusemide has the chemical name 4-chloro-N-furfuryl-5-sulfamoylanthranilic acid. It has the chemical formula C 12 H 11 ClN 2 O 5 S with a molecular weight of 330.7. The CAS number is 53-31.9. DESCRIPTION Frusemide is a white to off-white odourless crystalline powder. It is practically insoluble in water, sparingly soluble in alcohol, freely soluble in dilute alkali solutions and insoluble in dilute acids. Solutions for injection are prepared with sodium hydroxide giving solutions with a pH of 8.0 to 9.3. Frusemide should be protected from light. PHARMACOLOGY Frusemide is a potent diuretic with a rapid action. It inhibits sodium and chloride absorption in the ascending limb of Henle's loop and in both the proximal and distal tubules. The high degree of efficacy is due to this unique site of action. The action on the distal tubule is independent of any inhibitory effect on carbonic anhydrase or aldosterone. Frusemide may promote diuresis in cases which have previously proved resistant to other diuretics. Frusemide has no significant pharmacological effects other than on renal function. PHARMACOKINETICS Absorption Frusemide is rapidly absorbed from the gastrointestinal tract. Absorption rates in healthy subjects have been reported from 60 - 69 % and from 43 - 46 % in patients with end stage renal failure.
