WHAT GIAGRA IS USED
FOR
GIAGRA is indicated for
the
treatment
of
erectile
dysfunction,
which
is
the
inability to achieve or maintain
a penile erection sufficient for
satisfactory
sexual
performance.
In
order
for
GIAGRA
to
be
effective,
sexual stimulation is required.
HOW GIAGRA WORKS
It works by helping to relax the
blood
vessels
in
your
penis,
allowing
blood
to
flow
into
your
penis
when
you
get
sexually
excited.
GIAGRA
will only help
you
to
get
an
erection
if
you
are
sexually
stimulated.
You
should
not
take GIAGRA
if
you
do
not
have erectile
dysfunction.
BEFORE YOU USE GIAGRA

When you must not use it
DO NOT TAKE GIAGRA
•
If
you
are
taking
medicines
called
nitrates,
as the combination may
Cause
a
potentially
dangerous
decrease
in
your blood pressure. Tell
your
doctor
if
you
are
taking
any
of
these
medicines which are often
given for relief of angina
pectoris (or “chest pain”).
If you are not certain, ask
your doctor or pharmacist.
•
If you are using any of the
drugs
known
as
nitric
oxide donors such as amyl
nitrite (“poppers”) as the
combination may also lead
to a potentially dangerous
decrease
in
your
blood
pressure.
•
If
you
are
allergic
(hypersensitive)
to
GIAGRA
or
any
of
the
other
ingredients
of
GIAGRA.
•
If you have a severe heart
or liver problem.
•
If you have recently had a
stroke or a heart attack, or
if
you
have
low
blood
pressure.
•
If
you
have
certain
rare
inherited eye diseases
(such
as
retinitis
pigmentosa).
If you have ever had loss
of vision because of non-
arteritic anterior ischaemic

Each film coated tablets contains Sildenafil Citrate 35.12mg is

PHARMACODYNAMICS
Mechanism of action
The physiologic mechanism of erection of the penis involves release of
nitric oxide (NO) in the
corpus cavernosum during sexual stimulation. NO then activates the
enzyme guanylate cyclase,
which results in increased levels of cyclic guanosine monophosphate
(cGMP), producing smooth
muscle relaxation in the corpus cavernosum and allowing inow of
blood. Sildenal has no direct
relaxant effect on isolated human corpus cavernosum, but enhances the
effect of nitric oxide
(NO) by inhibiting phosphodiesterase type 5 (PDE5), which is
responsible for degradation of
cGMP in the corpus cavernosum.
When sexual stimulation causes local release of NO, inhibition of PDE5
by sildenal causes
increased levels of cGMP in the corpus cavernosum, resulting in smooth
muscle relaxation and
inow of blood to the corpus cavernosum. Sildenal at recommended
doses has no effect in the
absence of sexual stimulation.
_In vitro_, sildenal is found to be selective for PDE5. Its effect
is more potent on PDE5 than on other
known phosphodiesterases (10-fold for PDE5, > 80-fold for PDE1, >
