Date of revision WHAT DIAPO IS USED FOR This medicine is used: • to treat anxiety disorders • to control muscle spasm and relax muscles • to treat symptoms associated with alcohol withdrawal such as trembling and agitation • as hypnotic in short term management of insomnia • as an anticonvulsant for convulsions or seizures • to treat tension headache Ask your doctor if you have any questions about why this medicine has been prescribed for you. Your doctor may have prescribed this medicine for another reason. This medicine is available only with a doctor's prescription. HOW DIAPO WORKS This medicine belongs to a group of medicines called benzodiazepines. It is thought to work by acting on chemicals in the brain. Benzodiazepines act as depressants of the central nervous system (CNS) producing CNS depression from sedation to hypnosis to coma depending on dose. Benzodiazepines should not be used alone to treat depression or severe mental illness. In general, benzodiazepines such as diazepam should be taken for short periods only. Continuous long-term use is not recommended unless advised by your doctor. There is evidence that using these types of medicines can lead to dependence. BEFORE YOU TAKE DIAPO - _When you must not take it_ Do not take this medicine if: • you have severe lung disease • you suffer from sleep apnoea (temporary stops in breathing during sleep) • you suffer from myasthenia gravis (a disease which causes severe muscle weakness) • you are addicted to drugs or alcohol (unless your doctor has prescribed diazepam to help to relieve the symptoms of alcohol withdrawal) • the expiry date (EXP) printed on the pack has passed • the packaging is to Baca dokumen lengkap
DESCRIPTION DIAPO PHARMACOKINETICS INDICATIONS DOSAGE PHARMACODYNAMICS DIAPO TABLET 5MG A 8mm diameter, round, flat, scored yellow tablet. _Each tablet contains Diazepam 5mg_ Diazepam is a long-acting benzodiazepine with anticonvulsant, anxiolytic, sedative and muscle relaxant properties. It is indicated as an antianxiety agent, as a hypnotic in the short-term management of insomnia, as a sedative and preoperative medication, as an anticonvulsant particularly in the management of status epilepticus and febrile convulsions, in the control of muscle spasm as in tetanus, and in the management of alcohol withdrawal symptoms. It is also used as antipanic and antitremor agent. Diazepam is also used to relieve spasms of facial muscles associated with problems of occlusion and temporomandibular joint disorders and for the treatment of tension headache. In general, benzodiazepines act as depressants of the central nervous system producing all level of CNS depression from mild sedation to hypnosis to coma depending on the dosage. The precise sites and mechanisms of action have not been completely established. Although various mechanisms of action have been proposed, it is believed that diazepam enhance or facilitate the inhibitory neurotrans- mitter action of gamma-aminobutyric acid (GABA), which is one of the major inhibitory neurotransmitters in the brain and mediates both pre- and post-synaptic inhibition in all regions of the CNS, following interaction between the benzodiaze- pine and a specific neuronal membrane receptor. Diazepam reportedly act as agonists at the benzodiazepine receptors, which have been shown to form a component of a functional supramolecular unit known as the benzodiazepine-GA- BA receptor-chloride ionophore complex. The receptor complex, believed to reside on neuronal membranes that regulate cell firing, functions mainly in the gating of the chloride channel.
