Date of revision WHAT HP METFORMIN MR 850 MG TABLET IS USED FOR Used to treat type 2 diabetes or non- insulin dependent diabetes mellitus (NIDDM) in adults whose high blood sugar levels cannot be lowered through exercising or monitoring food intake. HOW HP METFORMIN MR 850 MG TABLET WORKS This medicine belongs to a group called biguanides that lowers the high blood sugar levels in the body. It works by: reducing the amount of blood sugar produced in the liver improving your body’s sensitivity towards insulin and the way the body uses insulin slowing down the rate of glucose being absorbed in the intestines. BEFORE YOU USE HP METFORMIN MR 850 MG TABLET _When you must not take it _ Do not take this medicine if: you are allergic to meformin, any other biguanide, or its excipients you have diabetic coma (when your blood sugar is abnormally high and body is severely dehydrated) you have severe burns and infections you have kidney problems you have liver problems you have undergone/are undergoing major surgery you have severe trauma you have an active or history of lactic acidosis (buildup of acid in the blood due to inefficient removal from the body) you have severe breathing difficulties you have heart or blood vessel problems eg. heart failure, heart attack, sudden/rapid development of blood failing to circulate in the body you are breastfeeding or plan to breastfeed you are pregnant or plan to become pregnant. _Before you start to take it _ Tell your doctor if: Baca dokumen lengkap
DESCRIPTION Round, white uncoated tablet with convex faces. COMPOSITION Metformin Hydrochloride 850 mg per tablet (Extended-Release) PHARMACODYNAMICS Metformin is an oral biguanide antidiabetic agent. Its mode of action is thought to be multifactorial and includes delayed uptake of glucose from the gastro-intestinal tract; increased peripheral glucose utilization mediated by increased insulin sensitivity; and inhibition of increased hepatic and renal gluconeogenesis. PHARMACOKINETICS Metformin Hydrochloride is slowly and incompletely absorbed from the gastrointestinal tract. The absolute bioavailability of a single 500mg dose is reported to be about 50 to 60%, although this is reduced somewhat if taken with food. Plasma protein binding is negligible. It is excreted unchanged in the urine. The plasma elimination half-life is reported to range from about 2 to 6 hours after oral administration. INDICATIONS Metformin is used in the treatment of non-insulin-dependent diabetes mellitus (type 2) in adults, not responding to exercise and dietary modification. Diabetmin may be used as monotherapy or in combination with other oral antidiabetic agents, or with insulin. CONTRAINDICATIONS This medication is contraindicated in patients with the following medical problems: • Hypersensitivity to metformin. • Any condition needing close blood glucose control, such as: severe burns, dehydration, diabetic coma, diabetic ketoacidosis, hyperosmolar nonketotic coma, severe infection, major surgery, and severe trauma. • Conditions associated with hypoxemia, such as: cardiorespiratory insufficiency, cardiovascular collapse, congestive heart failure, acute myocardial infarction. • Severe, acute, or chronic hepatic disease. • Active or history of lactic acidosis. • Renal function impairment or renal disease. • Diagnostic or medical examinations using intravascular iodinated contrast media such as: angiography, intravenous cholangiography, computed tomography (CT) scan, pyelography and urography. • Severely reduced kidney f Baca dokumen lengkap
