【臨床藥理】
作用機轉/效應：
Allopurinol
及其代謝物，Oxipurinol（Alloxanthine），會經由抑制
黃嘌呤氧化酶（Xanthine
oxidase）的作用而減少尿酸之生成。
黃嘌呤氧化酶是將次黃嘌呤（Hypoxanthine）轉換成黃嘌呤
（xanthine），再由黃嘌呤轉換成尿酸（Uric
Acid）。同時，
Allopurinol 也經由涉及 Hypoxanthine-guanine
phosphoribosyltransferase（HGPRTase）之作用而對核苷酸
（Nucleotide）與核酸（Nucleic
acid）的合成增加對次黃嘌呤與
黃嘌呤的再利用。核苷酸濃度增加的結果會導致嘌呤之重新合
成的回饋性抑制。因此，Allopurinol
乃同時減低血清以及尿液
中的尿酸濃度。
Allopurinol
由於同時減低血清與尿液中的尿酸濃度至低於其溶
解度限度而阻止或減低尿酸鹽（Urate）的沉著，因此，同時防
止痛風性關節炎（Gouty arthritis）與尿酸鹽腎病（Urate
nephropathy）的發生或進行。在有慢性痛風的病人，Allopurinol
會阻止或減低沙石（Tophi）的生成與慢性關節變化，促進既存
