Manufacturer and Product Registration Holder 10. Date of revision WHAT PORO ® TABLET IS USED FOR Poro ® Tablet is used for the relief of fever and mild to moderate pain. HOW PORO ® TABLET WORKS Poro ® Tablet consists of paracetamol, which is an analgesic that relieves pain and an antipyretic that reduces fever. BEFORE YOU USE PORO ® TABLET _- When you must not use it_ Do not use Poro ® Tablet if you have allergy towards paracetamol or any components in this preparation. _- Before you start to use it _ Tell your doctor or pharmacist you are allergic to paracetamol or you have any other health related problem. Inform your healthcare providers if you have suffered from kidney problems or liver diseases. _- Taking other medicines _ Do not take other paracetamol containing medicines at the same time. You must also inform your doctor or pharmacist about all the medicines you are taking, including those without a prescription. HOW TO USE PORO ® TABLET _- How much to use _ The usual adult dose is 1 – 2 tablets 3 – 4 times daily (Not more than 8 tablets per day unless prescribed by a doctor). Do not exceed the prescribed dose. _- When to use it _ Poro ® Tablet should be taken according to the time advised by your doctor or pharmacist. It can be taken with or without food. _- How long to use it _ Use Poro ® Tablet as directed by your doctor or pharmacist. If fever persists for more than 3 days or recurs, or if pain continues for more than 5 days, consult your doctor immediately. _- If you forget to use it _ If you missed a dose of PORO ® TABLET, take it as soon as possible. Paracetamol (Acetaminophen) may act by inhibiting prostaglandin synthesis in the central nervous system and through a peripheral action by blocking pain impulse generation. The peripheral action may also be due to inhibition of the synthesis of prostaglandins which sensitized pain receptors to mechani- cal or chemical stimulation. 2. Paracetamol produces antipyresis by acting centrally on the hypothalamic heat-regulation center to produce peripheral vasodilation resulting in increased blood flow through the skin, sweating and heat loss. The central action probably involves inhibition of prostaglandin synthesis in the hypothala- mus. 3. Paracetamol is readily and completely absorbed from the gastrointes- tinal tract with peak plasma concentration occurring between 15 minutes to 2 hours after ingestion. Paracetamol is uniformly distributed throughout most body fluids; the apparent volume of distribution is 1 to 1.2 L/kg. Paracetamol may cross placenta and excreted in milk. At therapeutic plasma levels, no protein binding occurs. Paracetamol is metabolized by the hepatic microsomal enzyme system. In adults at therapeutic doses, paracetamol is mainly conjugated with glucuro- nide (45-55%) or sulphate (20-30%). A minor proportion (less than 20%) is metabolized to catechol derivatives, and mercapturic acid compounds via oxidation. Paracetamol is metabolised differently by infants and children compared to adults, the sulphate conjugate being predominant. Paracetamol is excreted by active tubular secretion: 85-90% of the adminis- tered dose is eliminated in the urine within 24 hours of administration Paracetamol has a half-life of 1 to 4 hours. Less than 5 % is excreted as unchanged paracetamol. INDICATION(S): For the symptomatic relief of mild to moderate pain and to reduce fever.
