PHARMACOLOGY Carprofen is a non-steroidal antiinflammatory drug (NSAID) of the propionic acid class with antiinflammatory, antipyretic and analgesic activity. The mechanism of action of carprofen is not completely understood. As for other NSAIDs, carprofen is known to inhibit cyclooxygenase (COX), the enzyme responsible for the conversion of arachidonic acid into pro-inflammatory mediators. However carprofen is a relatively weak inhibitor of COX at therapeutic dose rates, suggesting other mechanisms may be at least partly responsible for its antiinflammatory activity. Carprofen has some inhibitory activity against phospholipase A 2 , although the clinical significance of this activity is uncertain. A mode of action not primarily involving inhibition of prostaglandin production may account for the improved safety profile of carprofen over other NSAIDs of the same class in terms of lower ulcerogenic potential in the gastrointestinal tract. Carprofen is rapidly and extensively absorbed following oral administration; bioavailability is greater than 90%. Carprofen has a low volume of distribution and, as for many NSAIDs, has a high level of plasma protein binding at greater than 98%. Carprofen metabolism in the dog occurs via glucuronidation and oxidative processes in the liver. Excretion of the metabolites occurs primarily (70-80%) via the faeces, with 10-20% eliminated in the urine. Some enterohepatic circulation occurs.
